In the last year of a Marsden-funded project investigating unusual post-translational modifications of a small antibacterial peptide, we made a breakthrough that showed this unique peptide was glycosylated not once but twice, and that the glycans were essential for its biological function. We have sequenced the bacteriocin gene cluster and would now like to investigate the enzyme(s) responsible for glycosylation.

This research project aims characterise the functions of these enzymes, and to prove that this novel bacteriocin is not alone; i.e. there are other bacteriocins similarly modified. Techniques used will include molecular biology, protein expression and purification, mass spectrometry, and x-ray crystallography.

The Institute of Molecular Biosciences has a strong research focus on cell and molecular biology with a vibrant interdisciplinary research culture. There are excellent research facilities available in the Institute including 'state of the art' facilities for microscopy (MMIC), DNA sequencing and structural biology. The Institute supports five Science majors: Biochemistry, Genetics, Plant Biology, Microbiology and Biological Sciences. It is located on the Manawatu campus of Massey University located in Palmerston North, New Zealand.

Qualifications.

Applicants should have recent PhD in biochemistry or a related discipline. Familiarity with the molecular biology and genetics of lactic acid bacteria is desirable, including skills in targeted gene disruption. Other skills sought are those in protein expression, especially in systems other than E. coli, protein chemistry and an interest in both structural biology and bioinformatics.
